Title: Synthesis and characterization of etherated amphiphiles: Part I. Etherated surfactants. Part II. Etherated phospholipids

Abstract: Part 1. A series of etherated surfactants were synthesized and characterized in order to assess the hydrophobicity/hydrophilicity of the ether group. These molecules have a typical hydrophilic head group (either a quaternary ammonium bromide or a sodium sulfate) but instead of the customary all-hydrocarbon tail, two ethers groups were randomly incorporated. The cationic surfactants were meticulously studied and compared to two standard quaternary ammonium surfactants (DTAB and TTAB). Characterization methods included HPLC, tensiometry, NMR, viscosity, cryo-etch HRSEM and so on. Among other things, it was found that the ether groups appear to induce morphological changes in the typical aggregation patterns. Additionally, ether oxygens seem to display a stronger effect when dispersed among a larger number of carbon units. Part II. Based on the intriguing results of the etherated surfactants ( Part One ), investigations into the effect of the ether group on bilayers were undertaken. To this end, the original synthesis of the etherated surfactants was extrapolated in order to obtain the desired phospholipids. Three of the four etherated lipids were then studied using 23 Na NMR to monitor their effects on sodium flux through vesicles. After careful, detailed investigations, an intriguing unprecedented result was discovered. Through analysis and integration of the data, a highly complex model system was proposed to explain these extraordinary findings.
