Title: DNA cleavage by complexes of the type CpMLnR and their application to studying DNA-protein interactions

Abstract: Due to their proven antitumor and antibiotic properties, and possible use as footprinting agents, compounds that cleave DNA are of interest. The ambient light photolysis of CpFe(CO) 2 Ph or Cp 2 Fe 2 (CO) 4 and H 2 O 2 in the presence DNA resulted in strand scission, a result that shows potential as a means of targeting tumors, due to the high levels of hydrogen peroxide in cancer cells. This cleavage process is dependent on the concentration of both CpFe(CO) 2 Ph or Cp 2 Fe 2 (CO) 4 and H 2 O 2 . In the case of CpFe(CO) 2 Ph, the mechanism of the cleavage appears to be both hydrolytic and oxidative. However, in the case of Cp 2 Fe 2 (CO) 4 , the process appears to be only hydrolytic. In addition, the spermine-[CpW(CO) 3 Ph] 2 conjugate cleaved DNA at a molecule to base pair (bp) ratio as low as 0.038 molecules bp -1 upon irradiation with light that is greater than 300 nm. In contrast to the histone modifying action of other complexes of the type CpMLnR, this compound selectively cleaves DNA in DNA-histone assemblies. Due to the potential advantages complexes of the type CpML n R as footprinting agents, initial experiments for the footprinting of unmodified nucleosome core particles and particles phosphorylated at serine 10 of histone H3 were completed.
